Excessive collagen accumulation is a hallmark of fibrosis pathogenesis.  In research and drug discovery, the most commonly used collagen quantization methods include quantification of hydroxyproline using HPLC, the evaluation of radio-labeled proline , or precipitation of collagen using Sirius Red.  These methods are not suitable for high throughput screening because they are time-consuming, expensive, and requirement of large quantity of cell sample.   

Scientists at Athena Discovery have developed proprietary technology to measure collagen production in cell culture; here are the advantages of this collagen assay method:

• TGF beta1 can induce collagen 1 production in dose dependent way
• Alk-5 inhibitor (SD208) at 0.25 uM can completely block TGF beta 1 induced collagen production  
• 96 well format, Reproducible
• Much easier than other methods, such as Sirius Red or hydroxyproline quantification, therefore produce very consistent results
• Collagen induction 12-24 hours, and collagen quantification 4 hours.
